      目的 观察“嗅三针”对帕金森病(Parkinsons disease,PD)小鼠黑质高迁移率组蛋白1(high mobility group box 1,HMGB1)及核因子-κB(nuclear factor-κB,NF-κB)蛋白表达水平的影响,探讨“嗅三针”改善PD的作用机制。方法 采用随机数字表法将健康雄性C57BL/6小鼠分为空白组、模型组、电针组、左旋多巴组,每组10只;除空白组外,其余每组采用脂多糖经鼻滴注制作小鼠PD模型。先采用Morris水迷宫试验对各组小鼠进行行为学检测,再采用免疫组织化学法检测小鼠黑质细胞中HMGB1的表达,蛋白免疫印迹法检测 HMGB1、NF-κB蛋白在黑质中的表达水平。结果 模型组小鼠抵达终点时间多于空白组(P<0.05);电针组到达终点时间较模型组明显减少(P<0.05)。与空白组比较,模型组黑质中HMGB1阳性细胞数升高(P<0.05),HMGB1、NF-κB蛋白表达水平升高(P<0.05);与模型组比较,电针组黑质中HMGB1阳性细胞数降低(P<0.05),电针组黑质中HMGB1表达水平下降(P<0.05)。结论 “嗅三针”可改善PD小鼠空间记忆能力,抑制PD小鼠神经炎症,这可能与其抑制黑质区HMGB1表达有关。

      Objective To investigate the effect of "Xiusanzhen" on the expression of high-mobility group box 1 (HMGB1) and nuclear factor-kappa B (NF-κB) in the substantia nigra of mice with Parkinsons disease (PD) and the mechanism of action of "Xiusanzhen" in improving PD. Methods A total of 40 healthy male C57BL/6 mice were divided into blank group, model group, electroacupuncture group, and levodopa group using a random number table, with 10 mice in each group, and all mice except those in the blank group were given intranasal instillation of lipopolysaccharide to establish a mouse model of PD. Morris water maze was used for behavioral analysis, immunohistochemistry was used to measure the expression of HMGB1 in the substantia nigra of mice, and Western blot was used to measure the protein expression of HMGB1 and NF-κB in the substantia nigra. Results Compared with the blank group, the model group had a significantly longer time to reach the destination (P<0.05), and compared with the model group, the electroacupuncture group had a significantly shorter time to reach the destination (P<0.05). Compared with the blank group, the model group had significant increases in the number of HMGB1-positive cells and the protein expression of HMGB1 and NF-κB in the substantia nigra (P<0.05), and compared with the model group, the electroacupuncture group had significant reductions in the number of HMGB1-positive cells and the protein expression of HMGB1 in the substantia nigra (P<0.05). Conclusion "Xiusanzhen" can improve spatial memory ability and inhibit neuroinflammation in PD mice, possibly by inhibiting the expression of HMGB1 in the substantia nigra.
