      目的 探究不同浓度的加味阳和汤对维甲酸灌胃、氢化可的松臀部肌肉注射造成的肾阳虚型骨质疏松症(osteoporosis,OP)模型大鼠骨密度(bone mineral density,BMD)及血清微量元素的影响。方法 将48只SD大鼠分为空白对照组,模型组,加味阳和汤低、中、高剂量组,阳性对照组,除空白对照组外其余均复制肾阳虚型OP模型鼠,分别给予1%羧甲基纤维素钠、加味阳和汤(5、10、20 g/kg)、戊酸雌二醇(estradiol valerate,E2V)(0.000 2 g/kg)灌胃,每日1次,持续灌胃6周。 取大鼠股骨及血清,观察大鼠股骨BMD、血清碱性磷酸酶(alkaline phosphatase,ALP)、抗酒石酸酸性磷酸酶(tartrate-resistant acid phosphatase,TRAP)〖JP〗水平及微量元素[镁(magnesium,Mg)、锌(zinc,Zn)、铁(ferrum,Fe)]的变化。结果 与模型组比较,加味阳和汤各剂量组大鼠BMD均有不同程度的上升,大鼠血清ALP、TRAP活性均显著降低(P<0.05),血清Fe、Zn水平均显著升高,差异有统计学意义(P<0.05),血清Mg水平的变化无统计学意义(P>0.05)。结论 加味阳和汤对肾阳虚型OP大鼠有明显的治疗作用,且其作用机制可能与调节血清微量元素水平,抑制骨的高转换率有关。

      Objective To investigate the effect of different concentrations of Jiawei Yanghe Decoction on bone mineral density (BMD) and serum trace elements in a rat model of osteoporosis (OP) with kidney Yang deficiency induced by intragastric administration of retinoic acid and gluteal intramuscular injection of hydrocortisone. Methods A total of 48 Sprague-Dawley rats were divided into blank control group, model group, low-, middle-, and high-dose Jiawei Yanghe Decoction groups, and positive control group. All rats except those in the blank control group were used to establish a model of OP with kidney Yang deficiency. The rats in the model group, low-, middle-, and high-dose Jiawei Yanghe Decoction groups, and positive control group were given 1% sodium carboxymethyl cellulose, Jiawei Yanghe Decoction (5, 10, and 20 g/kg), and estradiol valerate (0.0002 g/kg), respectively, by gavage, once a day for 6 consecutive weeks. The femur and serum were collected to measure BMD of the femur and serum levels of alkaline phosphatase (ALP), tartrate-resistant acid phosphatase (TRAP), and the trace elements magnesium (Mg), ferrum (Fe), and zinc (Zn). Results Compared with the model group, the low-, middle-, and high-dose Jiawei Yanghe Decoction groups had significant increases in BMD, significant reductions in serum ALP and TRAP, and significant increases in serum Fe and Zn (all P<0.05), while there was no significant difference in serum Mg level between the two groups (P>0.05). Conclusion Jiawei Yanghe Decoction exerts a marked therapeutic effect on OP rats with kidney Yang deficiency, possibly by regulating the serum levels of trace elements and inhibiting the high bone turnover rate.
