      目的 研究2型糖尿病人群常见体质类型阴虚质、痰湿质与白细胞介素 6(interleukin 6, IL 6)、白细胞介素 10(interleukin 10,IL 10)、白细胞介素 10受体1(interleukin 10 receptor 1,IL 10R1) 基因多态性的相关性,筛选2型糖尿病常见中医体质的部分易感基因,为2型糖尿病的早期防治提供一定的依据。方法 收集2型糖尿病患者共395例,进行基因检测,应用聚合酶链反应 限制性片段长度多态性(PCR RFLP)技术检测IL 6的rs1800796位点、IL 10的rs1800871位点、IL 10R1的rs2228055位点,进行基因分型。参照中华中医药学会《中医体质分类与判定》进行中医体质辨识。对2型糖尿病常见体质类型阴虚质、痰湿质与IL 6、IL 10、IL 10R1基因多态性的相关性进行统计分析。结果 痰湿质组IL 10 rs1800871 CT基因型比例明显低于非痰湿质CT基因型比例(30.4% vs 45.9%,χ2=6.137,P=0.013)。以IL 10 rs1800871 CC为参照,IL 10 rs1800871 CT为2型糖尿病阴虚质的主要危险因素(P=0.019,OR=2.593,95% CI为1.710~5.746);以IL 10 rs1800871 CT为参照,IL 10 rs1800871 TT为2型糖尿病痰湿质的主要危险因素(P=0.025,OR=1.809,95% CI为1.076~3.041)。结论 IL 10 rs1800871位点CT基因型是2型糖尿病阴虚质患者的危险因素,IL 10 rs1800871位点TT基因型是2型糖尿病痰湿质患者的危险因素,IL 10基因多态性可能是2型糖尿病阴虚质、痰湿质的分子遗传学基础之一。

      Objective To investigate the association of Yin deficiency constitution and phlegm dampness constitution with gene polymorphisms of interleukin 6 (IL 6), interleukin 10 (IL 10), and interleukin 10 receptor 1 (IL 10R1) in patients with type 2 diabetes, to screen out the susceptible genes of common traditional Chinese medicine (TCM) constitutions of type 2 diabetes, and to provide a basis for early prevention and treatment of type 2 diabetes. Methods A total of 395 patients with type 2 diabetes were enrolled and gene detection was performed. PCR RFLP was used to detect the genotypes of IL 6 rs1800796, IL 10 rs1800871, and IL 10R1 rs2228055. TCM constitution was determined according to Classification and Determination of TCM Constitution by China Association of Chinese Medicine. A statistical analysis was performed to investigate the association of Yin deficiency constitution and phlegm dampness constitution (two common types of constitution of type 2 diabetes) with gene polymorphisms of IL 6, IL 10, and IL 10R1. Results The phlegm dampness constitution group had a significantly lower proportion of the patients with CT genotype of IL 10 rs1800871 than the non phlegm dampness constitution group (30.4% vs 45.9%, χ2=6.137, P=0.013). With genotype CC of IL 10 rs1800871 as a control, CT genotype of IL 10 rs1800871 was a main risk factor for Yin deficiency constitution in patients with type 2 diabetes [odds ratio (OR)= 2.593, 95% confidence interval (CI): 1.710 5.746, P=0.019]; with CT genotype of IL 10 rs1800871 as a control, genotype TT of IL 10 rs1800871 was a main risk factor for phlegm dampness constitution in patients with type 2 diabetes (OR=1.809,95% CI: 1.076 3.041, P=0.025). Conclusion CT genotype of IL 10 rs1800871 is a risk factor for Yin deficiency constitution in patients with type 2 diabetes, and TT genotype of IL 10 rs1800871 is a risk factor for phlegm dampness constitution in patients with type 2 diabetes. IL 10 gene polymorphisms may be a molecular genetic basis for Yin deficiency constitution and phlegm dampness constitution in patients with type 2 diabetes.
