      目的 建立独活中香豆素类成分的高效液相色谱指纹图谱分析方法,为独活药材的质量控制提供依据。方法 采用C18 色谱柱(4.6 mm × 250 mm, 5 μm),以水-甲醇为流动相进行梯度洗脱,流速1.2 mL/min,柱温25 ℃,检测波长330 nm。结果 建立了独活药材香豆素类成分的高效液相色谱特征指纹图,标定了15个共有色谱峰,采用相似度评价、聚类分析、主成分分析对所收集的15批样品进行指纹图谱模式识别研究,对6个共有峰借助对照品进行了指认,采用高效液相色谱-电喷雾离子源-质谱技术并结合文献报道对其余9个共有峰进行了初步归属。15批独活药材的相似度结果为0.797~0.979。运用聚类分析,15批独活药材可大致聚成5类。结论 该法所建立的独活药材香豆素类成分指纹图谱特征性强,为该药材的质量控制提供了实验依据。

      Objective To develop a high-performance liquid chromatography (HPLC) method for the fingerprint analysis of coumarins from Radix Angelicae Pubescentis, and to provide a reference for its quality control. Methods HPLC was performed on a C18 column (4.6 mm × 250 mm, 5 μm) with a mobile phase of water-methanol for gradient elution at a flow rate of 1.2 mL/min, a column temperature of 25 ℃, and a detection wavelength of 330 nm. Results The HPLC characteristic fingerprints of coumarins from Radix Angelicae Pubescentis were established, and there were 15 common chromatographic peaks in the fingerprints. Fingerprint pattern recognition was performed on 15 batches of samples by similarity evaluation, cluster analysis, and principal component analysis. Among the 15 common peaks, 6 were confirmed with reference to the retention time (tR) and mass spectrum of the reference standard, and the remaining 9 peaks were preliminarily classified by HPLC-ESI-MSn as well as literature reference. The similarity of the fingerprints of 15 batches of samples was 0.797-0.979. The 15 batches of samples could be generally classified into 5 clusters using cluster analysis. Conclusion The fingerprints of coumarins from Radix Angelicae Pubescentis established by this method are highly characteristic, and may provide an experimental basis for the quality control of Radix Angelicae Pubescentis.
