      目的 观察参黄合剂对大鼠瘘管创面组织中血管内皮细胞信号通路FLK-1/KDR、PIP2、PIP3蛋白及其mRNA的影响。方法 将180只雄性SD大鼠按随机数字表法分为空白对照组、空白模型组、模型对照组、生理盐水组、高锰酸钾组、参黄合剂组,每组30只。采用弹簧纱条埋置于大鼠颈背部并加混合菌液的方法复制体表瘘管模型。术后l、7、14 d,分别采用Western blot法、RT-PCR法检测各组大鼠创面组织中FLK-1/KDR、PIP2、PIP3蛋白及其mRNA表达水平。结果 除空白对照组外,其余5组大鼠创面FLK-1/KDR、PIP2、PIP3蛋白及其mRNA表达水平均随术后时间的延长而明显升高;术后14 d,其余5组上述指标(除高锰酸钾组PIP2 mRNA外)表达水平显著高于术后1 d(P<0.05)。术后1、7、14 d,高锰酸钾组和参黄合剂组FLK-1/KDR、PIP2、PIP3蛋白及其mRNA表达水平均高于空白对照组、模型对照组和生理盐水组;术后7、14 d,参黄合剂组FLK-1/KDR、PIP2、PIP3蛋白及其mRNA表达水平均显著高于空白对照组、模型对照组和生理盐水组(P<0.05),部分指标表达水平显著高于高锰酸钾组(P<0.05)。结论 参黄合剂对大鼠瘘管创面组织中血管内皮细胞信号通路相关因子的表达具有促进作用,从而促进血管新生,加速创面愈合。

      Objective To investigate the effect of Shenhuang Mixture on the protein and mRNA expression of FLK-1/KDR, PIP2, and PIP3 in vascular endothelial cells in the wound tissue of fistula in rats. Methods A total of 180 male Sprague-Dawley rats were divided into blank control group, blank model group, model control group, normal saline group, potassium permanganate group, and Shenhuang Mixture group using a random number table, with 30 rats in each group. A rat model of surface fistula was established by embedding spring gauze at the back of the neck and adding a mixed bacterial solution. On days 1, 7, and 14 after surgery, Western blot and RT-PCR were used to measure the protein and mRNA expression of FLK-1/KDR, PIP2, and PIP3 in wound tissue. Results All groups except the blank control group had significant increases in the protein and mRNA expression of FLK-1/KDR, PIP2, and PIP3 in wound tissue over time after surgery; all indices except the mRNA expression of PIP2 in the potassium permanganate group increased significantly from day 1 to day 14 after surgery (P<0.05). On days 1, 7, and 14 after surgery, the potassium permanganate group and the Shenhuang Mixture group had significantly higher protein and mRNA expression of FLK-1/KDR, PIP2, and PIP3 than the blank control group, the model control group, and the normal saline group; on days 7 and 14 after surgery, the Shenhuang Mixture group had significantly higher protein and mRNA expression of FLK-1/KDR, PIP2, and PIP3 than the blank control group, the model control group, and the normal saline group (P<0.05), and some of these indices in the Shenhuang Mixture group were significantly higher than those in the potassium permanganate group (P<0.05). Conclusion Shenhuang Mixture can increase the expression of related factors of the signaling pathways in vascular endothelial cells in the wound of fistula in rats and thus promote angiogenesis and wound healing.
