Abstract:Objective To investigate the distribution characteristics of endophytic fungi in Dendrobium huoshanense from different habitats. Methods The DNA molecular calibration technique was used to identify the species of different Dendrobium huoshanense samples, and Green Shield Sequencing was used to analyze the distribution of endophytic fungi in Dendrobium huoshanense. Results All test samples were identified as Dendrobium huoshanense. The operational taxonomic unit (OTU) numbers of 10 samples of Dendrobium huoshanense (LP1, LP2, LD1, LD2, LS1, LS2, HP1, HP2, JP1, and JP2) were 540, 390, 405, 376, 121, 601, 423, 426, 392, and 394, respectively. Through an analysis of the species taxonomy of each OTU, the distribution of endophytic fungi in Dendrobium huoshanense could be attributed to the species level, and the numbers of the species of endophytic fungi in these samples were 140, 110, 134, 128, 133, 142, 144, 127, 151, and 135, respectively. The OTUs shared by LP1 and LP2, LD1 and LD2, LS1 and LS2, HP1 and HP2, and JP1 and JP2 accounted for 84.22%, 95.58%, 94.52%, 93.59%, and 94.05%, respectively, on the species level. Conclusion There are differences in the species of endophytic fungi between Dendrobium huoshanense from different habitats. Dendrobium huoshanense samples from the same habitat have a similar community composition of endophytic fungi, and the growth of endophytic fungi in Dendrobium huoshanense has regional specificity.
