养阴活胃合剂对慢性萎缩性胃炎大鼠血清胃蛋白酶原和胃组织水通道蛋白的影响

    摘要:

    目的 探究养阴活胃合剂治疗慢性萎缩性胃炎(chronic atrophic gastritis,CAG)的作用机制。方法 将66只SD大鼠随机分为正常组,模型组,胃复春组,养阴活胃合剂高、中、低剂量组。采用综合法(氨水+脱氧胆酸钠+不同浓度乙醇+饥饱失常法)复制CAG模型,连续12周。采用ELISA法检测大鼠血清胃蛋白酶原Ⅰ(pepsinogen Ⅰ,PGⅠ)、胃蛋白酶原Ⅱ(pepsinogen Ⅱ,PGⅡ)水平及胃蛋白酶原比值(pepsinogen ratio,PGR)。采用RT-qPCR法检测胃组织水通道蛋白(aquaporin,AQP)1、AQP3和AQP4 mRNA表达水平。〖JP〗结果 与模型组比较,养阴活胃合剂高、中、低剂量及胃复春可以提高大鼠胃液总酸度,改善胃液pH值(P<0.05);各给药组大鼠血清PGⅠ水平和PGR升高,PGⅡ水平降低;各给药组大鼠AQP1 mRNA表达水平降低,养阴活胃合剂中、低剂量组与胃复春组大鼠AQP3 mRNA表达水平明显升高(P<0.05),养阴活胃合剂高、中剂量组和胃复春组AQP4 mRNA表达水平显著升高(P<0.05)。结论 养阴活胃合剂通过升高血清PGⅠ水平和PGR,上调AQP3、AQP4 mRNA表达水平,降低PGⅡ水平和下调AQP1 mRNA表达水平改善CAG大鼠的胃酸分泌。

    Abstract:

    Objective To investigate the mechanism of action of Yangyin Huowei Mixture in the treatment of chronic atrophic gastritis (CAG). Methods 〖JP2〗A total of 66 Sprague-Dawley rats were randomly divided into normal group, model group, Weifuchun group, and high-, middle-, 〖JP〗and low-dose Yangyin Huowei Mixture groups. A comprehensive method (ammonia+sodium deoxycholate+different concentrations of ethanol+irregular diet) was used to establish a model of CAG for 12 consecutive weeks. ELISA was used to measure the levels of pepsinogen Ⅰ (PGⅠ), pepsinogen Ⅱ (PGⅡ), and pepsinogen Ⅰ/Ⅱ ratio (PGR), and RT-qPCR was used to measure the mRNA expression levels of aquaporin-1 (AQP1), aquaporin-3(AQP3), and aquaporin-4 (AQP4) in gastric tissue. Results Compared with the model group, the high-, middle-, and low-dose Yangyin Huowei Mixture groups had a significant increase in total acidity of gastric juice and a significant improvement of the pH value of gastric juice (P<0.05). All administration groups had increases in the serum levels of PGⅠ and PGR and a reduction in the serum level of PGⅡ. All administration groups had a reduction in the mRNA expression level of AQP1; the middle- and low-dose Yangyin Huowei Mixture groups and the Weifuchun group had a significant increase in the mRNA expression level of AQP3 (P<0.05), and the high- and middle-dose Yangyin Huowei Mixture groups and the Weifuchun group had a significant increase in the mRNA expression level of AQP4 (P<0.05). Conclusion Yangyin Huowei Mixture can improve the secretion of gastric acid in CAG rats by increasing the serum levels of PGⅠ and PGR, upregulating the mRNA expression levels of AQP3 and AQP4, and downregulating the mRNA expression levels of PGⅡ and AQP1.
