电针太冲、足三里对自发性高血压大鼠心肌纤维化的改善作用

    目的 观察电针对于自发性高血压大鼠(spontaneously hypertensive rats,SHR)收缩压及左心室血管紧张素Ⅱ(angiotensin Ⅱ,AngⅡ),细胞外信号调节激酶1/2(extracellular-regulated kinase 1/2,ERK1/2),胶原蛋白Ⅰ、胶原蛋白Ⅲ含量的影响,探讨电针抑制高血压心肌纤维化的作用机制。方法 将36只SHR随机分为模型组、电针组、药物组,每组12只,另设12只Wistar大鼠作为空白组。模型组和空白组只进行抓取和固定刺激,电针组大鼠选取双侧太冲、足三里穴进行电针刺激,药物组服用氯沙坦钾。实验周期21 d。实验结束后测量大鼠尾动脉收缩压,Masson染色观察大鼠左心室病理改变,酶联免疫吸附法检测大鼠左心室AngⅡ含量,qPCR检测大鼠左心室ERK1/2、胶原蛋白Ⅰ、胶原蛋白Ⅲ mRNA表达水平。结果 染色结果显示,电针组及空白组左心室心肌组织形态正常,模型组心肌组织间大量胶原纤维增生,药物组心肌组织间可见少量胶原纤维。与空白组比较,各时点模型组大鼠尾动脉收缩压均显著升高(P<0.05);与模型组比较,治疗第14天、第21天电针组、药物组收缩压显著降低(P<0.05)。与空白组比较,模型组大鼠左心室AngⅡ含量,ERK1/2、胶原蛋白Ⅰ、胶原蛋白Ⅲ mRNA表达水平均显著升高(P<0.05);与模型组比较,电针组、药物组大鼠左心室AngⅡ含量,ERK1/2、胶原蛋白Ⅰ、胶原蛋白Ⅲ mRNA表达水平均显著降低(P<0.05)。结论 电针能够对AngⅡ、ERK信号通路产生影响,进而下调大鼠心肌胶原蛋白mRNA的表达,从而起到抑制高血压大鼠心肌纤维化的作用,这可能是针刺能够有效减缓高血压心脏损伤的机制之一。

    Abstract:

    Objective To investigate the effect of electroacupuncture on systolic pressure and the content of angiotensin Ⅱ (AngⅡ), extracellular signal-regulated kinase 1/2 (ERK1/2), collagen type Ⅰ, and collagen type Ⅲ in spontaneously hypertensive rats (SHRs), as well as the mechanism of action of electroacupuncture in inhibiting myocardial fibrosis in the presence of hypertension. Methods A total of 36 SHRs were randomly divided into model group, electroacupuncture group, and drug group, with 12 rats in each group, and 12 Wistar rats were selected as blank control group. The rats in the model group and the blank group were captured, fixed, and stimulated, those in the electroacupuncture group were given electroacupuncture at Taichong and Zusanli on both sides, and those in the drug group were given losartan potassium. The duration of the experiment was 21 days. Systolic pressure of the caudal artery was measured after experiment; Masson staining was used to observe the pathological changes of the left ventricle; ELISA was used to measure the content of AngⅡ in the left ventricle; qPCR was used to measure the mRNA expression levels of ERK1/2, collagen type Ⅰ, and collagen type Ⅲ in the left ventricle. Results Staining showed that the electroacupuncture group and the blank control group had normal histomorphology of the left ventricle, the model group had proliferation of a large amount of collagen fibers in the myocardial tissue, and the drug group had a small amount of collagen fibers in the myocardial tissue. Compared with the blank control group, the model group had a significant increase in the systolic pressure of the caudal artery at each time point (P<0.05), and compared with the model group, the electroacupuncture group and the drug group had a significant reduction in systolic pressure on days 14 and 21 of treatment (P<0.05). Compared with the blank control group, the model group had significant increases in the content of AngⅡ and the mRNA expression levels of ERK1/2, collagen type Ⅰ, and collagen type Ⅲ in the left ventricle (P<0.05), and compared with the model group, the electroacupuncture group and the drug group had significant reductions in the content of AngⅡ and the mRNA expression levels of ERK1/2, collagen type Ⅰ, and collagen type Ⅲ in the left ventricle (P<0.05). Conclusion Electroacupuncture can inhibit myocardial fibrosis in hypertensive rats by affecting the AngⅡ and ERK signaling pathways and downregulating the mRNA expression of collagen in the myocardium, which may be one of the mechanisms of acupuncture in effectively alleviating cardiac injury induced by hypertension.
