    摘要:

    目的 观察阳和平喘颗粒对慢性哮喘气道重塑的影响,并探究其作用机制。方法 将50只大鼠随机分为正常组、模型组、地塞米松组、阳和平喘颗粒低剂量组和阳和平喘颗粒高剂量组,每组10只;采用卵清蛋白致敏激发复制哮喘大鼠模型。苏木素-伊红染色观察支气管肺组织病理变化;ELISA法检测肺泡灌洗液(bronchoalveolar lavage fluid, BALF)中白细胞介素4(interleukin 4, IL-4)、白细胞介素6(interleulin 6, IL-6 )水平;Western blot法检测支气管肺组织中转化生长因子β1(transforming growth factor-beta 1, TGF-β1)、Smad2与Smad3蛋白相对表达水平。结果 与正常组比较,模型组支气管管腔变窄,平滑肌层及基底膜层增厚,周围可见炎症细胞浸润,BALF中IL-4、IL-6水平升高(P<0.05),支气管肺组织中TGF-β1、Smad2与Smad3蛋白相对表达水平升高(P<0.05);与模型组比较,阳和平喘颗粒高、低剂量组和地塞米松组大鼠支气管肺组织病理学改变较轻, BALF中IL-4、IL-6水平下降(P<0.05),支气管肺组织中TGF-β1、Smad2与Smad3蛋白相对表达水平降低(P<0.05),与阳和平喘颗粒低剂量组比较,阳和平喘颗粒高剂量组和地塞米松组差异显著(P<0.05)。结论 阳和平喘颗粒可以改善慢性哮喘大鼠的气道重塑,减轻气道炎症,其机制可能与调节TGF-β1/Smad信号通路有关。

    Abstract:

    Objective To investigate the effect of Yanghe Pingchuan Granule on airway remodeling in rats with chronic asthma and related mechanism. Methods A total of 50 rats were randomly divided into normal group, model group, dexamethasone group, and low- and high-dose Yanghe Pingchuan Granule groups, with 10 rats in each group, and ovalbumin sensitization and challenge was used to establish a rat model of asthma. HE staining was used to observe the pathological changes of lung tissue; ELISA was used to measure the levels of interleukin 4 (IL-4) and interleukin-6 (IL-6) in bronchoalveolar lavage fluid (BALF); Western blot was used to measure the relative protein expression of transforming growth factor-beta 1 (TGF-β1), Smad2, and Smad3 in bronchial and lung tissue. Results Compared with the normal group, the model group had narrowing of the bronchial cavity, thickening of smooth muscle and basilar membrane, inflammatory cell infiltration, and increases in the levels of IL-4 and IL-6 in BALF (P<0.05), as well as significant increases in the relative protein expression of TGF-β1,Smad2, and Smad3 in bronchial and lung tissue (P<0.05). Compared with the model group, the high- and low-dose Yanghe Pingchuan Granule groups and the dexamethasone group had significant improvement in the pathological changes of lung tissue and significant reductions in the levels of IL-4 and IL-6 in BALF (P<0.05), as well as significant reductions in the relative protein expression of TGF-β1,Smad2, and Smad3 in bronchial and lung tissue (P<0.05). There were significant differences in these indices between the low-dose Yanghe Pingchuan Granule group and the high-dose Yanghe Pingchuan Granule group and dexamethasone group (P<0.05). Conclusion Yanghe Pingchuan Granule can improve airway remodeling and alleviate airway inflammation in rats with chronic asthma, possibly by regulating the TGF-β1/Smad signaling pathway.
