    摘要:

    目的 探讨左归饮水提液延缓肾阴虚大鼠衰老的机制。方法 将SD大鼠随机分为5组,分别为正常组,模型组,左归饮高、中、低剂量组,模型组每日灌胃给予甲状腺素混悬液150 mg/kg,左归饮高、中、低剂量组分别灌胃左归饮水提液31、20.67、13.78 g/kg,连续7 d。检测大鼠血清超氧化物歧化酶(superoxide dismutase, SOD)、丙二醛(malondialdehyde, MDA)、过氧化氢酶(catalase, CAT)、谷胱甘肽过氧化物酶(glutathione peroxidase, GSH-Px)、肌酐(creatinine, Cr)、血尿素氮(blood urea nitrogen, BUN)、环磷酸腺苷(cyclic adenosine monophosphate, cAMP)、环磷酸鸟苷(cyclic guanosine monophosphate, cGMP)水平及cAMP/cGMP比值。结果 与模型组比较,左归饮高、中、低剂量组大鼠血清cAMP水平,cAMP/cGMP均显著降低(P<0.05),而cGMP水平无显著变化;与模型组比较,左归饮中、高剂量组大鼠血清SOD、CAT、GSH-Px水平均显著升高(P<0.05),MDA水平显著下降(P<0.05);与模型组比较,左归饮低、中、高剂量组大鼠血清Cr和BUN水平均显著降低(P<0.05)。结论 左归饮通过抗氧化作用延缓肾阴虚大鼠衰老。

    Abstract:

    Objective To investigate the mechanism of water extract of Zuogui Decoction in delaying senescence of rats with kidney Yin deficiency. Methods Sprague-Dawley rats were randomly divided into normal group, model group, and high-, middle-, and low-dose Zuogui Decoction groups. The rats in the model group were given thyroxine suspension 150 mg/kg every day by gavage, and those in the high-, middle-, and low-dose Zuogui Decoction groups were given water extract of Zuogui Decoction at a dose of 31, 20.67, and 13.78 g/(kg·d), respectively, by gavage. The course of treatment was 7 consecutive days. The levels of superoxide dismutase (SOD), malondialdehyde (MDA), catalase (CAT), glutathione peroxidase (GSH-Px), creatinine (Cr), blood urea nitrogen (BUN), cyclic adenosine monophosphate (cAMP), and cyclic guanosine monophosphate (cGMP) were measured, and cAMP/cGMP ratio was calculated. Results Compared with the model group, the high-, middle-, and low-dose Zuogui Decoction groups had a significant reduction in the serum level of cAMP (P<0.05); the three Zuogui Decoction groups had no significant change in the level of cGMP, with a significant reduction in cAMP/cGMP ratio (P<0.05). Compared with the model group, the middle- and high-dose Zuogui Decoction groups had significant increases in the serum levels of SOD, CAT, and GSH-Px (P<0.05) and a significant reduction in MDA level (P<0.05). Compared with the model group, the low-, middle-, and high-dose Zuogui Decoction groups had significant reductions in the serum levels of Cr and BUN (P<0.05). Conclusion Zuogui Decoction can delay senescence of rats with kidney Yin deficiency by exerting an antioxidant effect.
