参七虫草胶囊干预肺间质纤维化大鼠模型PI3K/Akt信号通路实验研究

    摘要:

    目的 观察参七虫草胶囊对肺间质纤维化模型大鼠磷脂酰肌醇3激酶(phosphatidylinositol 3-kinase,PI3K)和丝氨酸/苏氨酸蛋白激酶(serine/threonine protein kinase B,Akt)信号通路的干预作用。方法 采用博莱霉素气道喷雾法复制大鼠肺间质纤维化模型。将40只大鼠随机分为空白对照组、模型组、氢化泼尼松组、参七虫草胶囊组,每组10只。酶联免疫吸附法测定各组大鼠治疗前后血清及肺组织中PI3K/Akt水平。结果 与空白对照组比较,模型组大鼠血清和肺组织中Akt及PI3K水平显著升高(P<0.05);与模型组比较,参七虫草胶囊组和氢化泼尼松组大鼠血清及肺组织中Akt及PI3K水平显著降低(P<0.05);参七虫草胶囊组大鼠血清和肺组织中Akt、PI3K水平显著低于氢化泼尼松组(P<0.05)。结论 参七虫草胶囊通过抑制PI3K/Akt信号通路的激活从而抑制肺纤维化炎症反应,延缓肺间质纤维化病程进展。

    Abstract:

    Objective To investigate the intervention effect of Shenqichongcao Capsule on the PI3K/Akt signaling pathway in a rat model of pulmonary interstitial fibrosis. Methods A rat model of pulmonary interstitial fibrosis was established by bleomycin airway spraying. A total of 40 rats were randomly divided into blank control group, model group, hydroprednisone group, and Shenqichongcao Capsule group, with 10 rats in each group. ELISA was used to measure the levels of phosphatidylinositol 3-kinase (PI3K) and serine/threonine protein kinase B (Akt) in 〖JP3〗serum and lung tissue before and after treatment. Results Compared with the blank control group, the model group had significant increases in the levels of Akt and PI3K in serum and lung tissue (P<0.05). Compared with the model group, the Shenqichongcao Capsule group and the hydroprednisone group had significant reductions in the levels of Akt and PI3K in serum and lung tissue (P<0.05), and the Shenqichongcao Capsule group had significantly lower levels than the hydroprednisone group (P<0.05). Conclusion Shenqichongcao Capsule can inhibit pulmonary fibrotic inflammatory response and delay the progression of pulmonary interstitial fibrosis by inhibiting the PI3K/Akt signaling pathway.
