目的 观察补益肺肾汤对变应性哮喘大鼠模型辅助性T淋巴细胞（helper T cell,Th）1/Th2的影响，探讨其治疗变应性哮喘的作用机制。方法 选取60只雄性SD大鼠，将其随机分成正常对照组、模型组、生理盐水组、补益肺肾组，每组15只，采用0.5%磷酸组胺溶液超声雾化法复制变应性哮喘大鼠模型，最后一次激发后处死大鼠，收集血清和支气管肺泡灌洗液（broncho alveolar lavage fluid，BALF），采用酶联免疫吸附法检测大鼠血清免疫球蛋白E（immunoglobin E，IgE）、白细胞介素-4（interleukin-4，IL-4）和干扰素γ（interferon-gamma, IFN-γ）水平，用流式细胞仪检测脾Th1、Th2细胞比例。结果 与正常对照组比较，模型组大鼠血清IgE和IL-4水平均显著升高（P＜0.05）；与模型组比较，补益肺肾组血清IgE和IL-4水平均显著降低（P＜0.05）；补益肺肾组大鼠血清IgE和IL-4水平显著低于生理盐水组（P＜0.05）。与模型组比较，生理盐水组和补益肺肾组大鼠BALF中IL-4水平明显降低（P＜0.05），IFN-γ水平明显上升（P＜0.05）；补肾益肺组大鼠BALF中IFN-γ水平显著高于生理盐水组（P＜0.05）。与模型组比较，生理盐水组和补益肺肾组大鼠脾中Th1/Th2明显上升（P＜0.05）；补肾益肺组大鼠脾中Th1/Th2显著高于生理盐水组（P＜0.05）。结论 补益肺肾汤可能通过下调IL-4水平和提高IFN-γ水平，调节变应性哮喘大鼠体内Th1/Th2失衡。
Objective To investigate the effect of Buyi Feishen Decoction on T-helper type 1 (Th1)/T-helper type 2 (Th2) balance in a rat model of allergic asthma and its mechanism of action in the treatment of allergic asthma. Methods A total of 60 male Sprague-Dawley rats were randomly divided into normal control group, model group, normal saline group, and Buyi Feishen Decoction group, with 15 rats in each group. Ultrasonic atomization of 0.5% histamine phosphate solution was used to establish a rat model of allergic asthma. The rats were sacrificed after the last challenge, and serum samples and bronchoalveolar lavage fluid (BALF) were collected. Enzyme-linked immunosorbent assay was used to measure the serum levels of immunoglobin E (IgE), interleukin-4 (IL-4), and interferon-gamma (IFN-γ), and flow cytometry was used to measure the percentages of Th1 and Th2 cells in the spleen. Results Compared with the model group, the Buyi Feishen Decoction group had significant reductions in the serum levels of IgE and IL-4 (P<0.05), and the Buyi Feishen Decoction group had significantly lower serum levels of IgE and IL-4 than the normal saline group (P<0.05). Compared with the model group, the normal saline group and the Buyi Feishen Decoction group had a significant reduction in the level of IL-4 and a significant increase in the level of IFN-γ in BALF (P<0.05), and the Buyi Feishen Decoction group had a significantly higher level of IFN-γ in BALF than the normal saline group (P<0.05). Compared with the model group, the normal saline group and the Buyi Feishen Decoction group had a significant increase in Th1/Th2 ratio in the spleen (P<0.05), and the Buyi Feishen Decoction group had a significantly higher Th1/Th2 ratio in the spleen than the normal saline group (P<0.05). Conclusion Buyi Feishen Decoction can regulate Th1/Th2 imbalance in rats with allergic asthma, possibly by downregulating IL-4 and upregulating IFN-γ.
