目的 探讨加减薯蓣丸对阿尔茨海默病(Alzheimers disease，AD)模型大鼠海马区髓细胞触发受体2(triggering receptor expressed on moyeloid cells 2,TREM2)、Iba1蛋白表达水平及对Iba1+细胞数量的影响。方法 采用SD大鼠双侧侧脑室注射β淀粉样蛋白1-42建立AD模型，将模型复制成功的大鼠分为模型组、中药组，并设正常组进行对照，每组10只。正常组、模型组大鼠每日给予10 mL/kg生理盐水灌胃，中药组大鼠每日给予加减薯蓣丸10 g/kg灌胃，连续灌胃4周后，应用Western blot法检测海马区TREM2和Iba1蛋白表达水平，免疫荧光染色标记Iba1+细胞数量。结果 与正常组比较，模型组TREM2蛋白表达水平显著增加（P＜0.05）；与模型组比较，中药组TREM2蛋白表达水平显著降低（P＜0.05），Iba1蛋白表达水平显著增加（P＜0.05），中药组海马区Iba1+细胞数量明显增加（P＜0.05）。结论 加减薯蓣丸可能通过影响海马区TREM2及Iba1蛋白水平的表达，增加Iba1+细胞数量，发挥对AD的治疗作用。
Objective To investigate the effect of modified Dioscorea opposita Thunb. Pill on the protein expression of triggering receptor expressed on myeloid cells 2 (TREM2) and Iba1 and the number of Iba1-positive cells in the hippocampus of rats with Alzheimers disease (AD). Methods Sprague-Dawley rats were given intracerebroventricular injection of beta-amyloid (1-42) at both sides to establish a model of AD, and after the model was successfully established, the rats were divided into model group and traditional Chinese medicine (TCM) group, with 10 rats in each group. A total of 10 normal rats were selected as normal group for control. The rats in the normal group and the model group were given 10 mL/kg normal saline by gavage every day, and those in the TCM group were given 10 g/kg modified Dioscorea opposita Thunb. Pill by gavage every day. After 4 consecutive weeks of treatment by gavage, Western blot was used to measure the protein expression of TREM2 and Iba1 in the hippocampus, and immunofluorescent staining was used to measure the number of Iba1-positive cells. Results Compared with the normal group, the model group had a significant increase in the protein expression of TREM2 (P<0.05), and compared with the model group, the TCM group had a significant reduction in the protein expression of TREM2 (P<0.05). There was a significant increase in the protein expression of Iba1 (P<0.05). Compared with the model group, the TCM group had a significant increase in the number of Iba1-positive cells(P<0.05). Conclusion Modified Dioscorea opposita Thunb. Pill exerts a therapeutic effect on AD by influencing the protein expression of TREM2 and Iba1 and increasing the number of Iba1-positive cells in the hippocampus.
